P-22: Effect of Bovine Cumulus Cell Co-Cultureon Maturation of Buffalo (Bubalus Bubalis)Oocyte

Background: The buffalo (Bubalus bubalis) is the animal with special production characteristics in producing milk and meat has a critical role in rural industry in Iran. The most problem of this specie is its low reproductive output. The aim of the present study was to evaluate the effect of bovine cumulus co-culture on maturation rate of Buffalo oocyte.Materials and Methods: Ovaries were collected from slaughtered buffaloes. The cumulus oocyte complexes (COC) were picked up form follicles (2-8 mm). The grade A and B COCs were randomly divided into 3 groups. Group 1; COCs were subjected to oocyte maturation medium containing TCM-199+ 5%FCS + 5%FF + 10μg/ml LH + 10 μg/ml FSH. Group 2; the COCs were cultured on a monolayer culture of bovine cumulus cells in a medium contained TCM199 with 5% FCS. Group 3; the denuded COCs were subjected on a monolayer culture of bovine cumulus cells in a medium similar to group 2. After 24 h of maturation in an atmosphere with 5 % CO2 and 95% relative humidity, the nuclear maturation of the oocytes was evaluated using a conventional aceto- orcein stain. Results: The maturation rate was significantly (p0.05). Conclusion: The results of the present study indicate the high efficacy of bovine cumulus cell monolayer on maturation of buffalo oocytes.
